Kisspeptin-10: KISS1R and neuroendocrine signaling in research

Kisspeptin-10 research peptide – effects, uses and comparison

Kisspeptin-10, Also known as KP-10, it is a C-terminal decapeptide derived from the products of the KISS1 gene. In scientific research, it is primarily studied in relation to the KISS1R receptor, neuroendocrine signaling, GnRH regulation, and the experimental evaluation of hypothalamic-pituitary mechanisms.

When interpreting the available data, it is important to distinguish between: endogenous kisspeptins, Kisspeptin-10, Kisspeptin-54, synthetic analogs KISS1R and specific research-grade material. Results obtained for one molecular form or a specific clinical protocol cannot be automatically transferred to another form or a commercial laboratory batch.

In short: Kisspeptin-10 is a 10-amino acid peptide with the sequence YNWNSFGLRF-NH₂. It activates the KISS1R receptor, historically referred to as GPR54. Experimental and clinical research mainly examines its relationship to GnRH/LH signaling and the dynamics of the neuroendocrine system. The results of these studies do not constitute a guide for use or evidence of the safety of a specific research-grade material.

What is Kisspeptin-10?

Kisspeptin-10 is a short peptide fragment derived from the C-terminal part of human kisspeptin. The term is also used in the professional literature. KP-10, kisspeptin-112–121 or human metastin 45–54.

The human sequence of Kisspeptin-10 is:

YNWNSFGLRF-NH₂

It is a C-terminal amidated decapeptide. The PubChem database for human Kisspeptin-10 lists the molecular formula C₆₃H₈₃N₁₇O₁₄, a relative molecular weight of approximately 1302.4 g/mol, and CAS 374675-21-5.

KISS1 and the KISS1R receptor

Gene KISS1 It encodes a precursor from which peptide products are formed, collectively referred to as kisspeptins. Their biological signaling occurs primarily through the receptor. KISS1R, which in older literature was referred to as GPR54.

KISS1R is among the G protein receptors and the research on this system has significantly contributed to the understanding of the regulation of GnRH neurons and neuroendocrine mechanisms related to hypothalamic-pituitary signaling.

Therefore, when searching for literature, it is important to use both designations:

  • KISS1R,
  • GPR54.

An older publication may use GPR54, although it describes the receptor today commonly referred to as KISS1R.

Receptor signaling KISS1R

KISS1R is a G protein receptor associated primarily with Gαq/11 signaling. Experimental cell work investigated the activation of phospholipase C, protein kinase C, and changes in intracellular Ca²⁺ following activation by kisspeptin receptor.

One of the mechanistic studies using cells expressing KISS1R showed that exposure to Kisspeptin-10 can lead to prolonged receptor signaling and Ca²⁺ mobilization in this model system.

Primary source:

Single-cell analyses of sustained kisspeptin-induced KISS1R signaling.

Interpretation:

Receptor activation, Ca²⁺ mobilization, or changes in downstream signaling represent mechanistic data. By themselves, they do not demonstrate clinical benefit, safety, or suitability for specific human use.

GnRH and LH in research models

The KISS1/KISS1R system is studied primarily as an upstream regulator gonadotropin-releasing hormone (GnRH) signalling.

GnRH subsequently influences the secretion of gonadotropins from the pituitary gland, with luteinizing hormone (LH) often used in research protocols as an indirect marker of the dynamics of GnRH signaling.

However, it is important to distinguish between the different levels:

Level Research significance
KISS1 / kisspeptin The ligand system studied as part of neuroendocrine regulation.
KISS1R G protein-coupled kisspeptin receptor.
GnRH Neuroendocrine signaling downstream of the KISS1/KISS1R system.
LH / FSH Hypophysial hormones used as measurable parameters in some research protocols.

What did the human mechanistic studies show?

Unlike many experimental peptides, there are also controlled physiological studies on Kisspeptin-10 in humans.

In a study published in 2011, researchers examined healthy men and evaluated changes in LH secretion after exposure to Kisspeptin-10. They observed changes in LH concentration and the dynamics of its pulses.

Primary source:

George et al. – Kisspeptin-10 is a potent stimulator of LH and increases pulse frequency in men.

Another study examining healthy men showed that exposure to C-terminal decapyquinone kisspeptin can affect the timing of the LH pulse used as an indirect indicator of GnRH signaling.

Kisspeptin resets the hypothalamic GnRH clock in men.

These studies provide data on human neuroendocrine physiology. However, they do not mean that the research-grade product is clinically approved or that the results of a particular protocol can be transferred outside of a controlled research environment.

Research in women and the importance of the experimental context

The response of the KISS1/KISS1R system is not identical under all biological conditions.

For example, a study in healthy women evaluated the response to kisspeptin at different phases of the menstrual cycle. The authors observed differences in the LH response according to the phase of the cycle.

Primary source:

Administration of Kisspeptin to Women: GnRH Response Across the Menstrual Cycle.

This result is a good example of why it is not appropriate to describe Kisspeptin-10 in a single universal sentence such as „increases hormones“. The resulting response depends on the biological context, population, protocol, and endpoint being monitored.

Genetic evidence for the KISS1/KISS1R system

Human genetic data also play an important role in the function of KISS1/KISS1R.

Publication in New England Journal of Medicine She described the inactivating mutation of the gene from 2012 KISS1 in a family with a disorder of normal pubertal progression.

Primary source:

Topaloglu et al. – Inactivating the KISS1 mutation and hypogonadotropic hypogonadism.

Genetic studies also show the importance of the KISS1R/GPR54 receptor for the normal functioning of this neuroendocrine axis.

Genetic evidence of the importance of the natural system is not evidence of the clinical safety or efficacy of exogenous synthetic Kisspeptin-10.

More recent data from 2026

In 2026, another mechanistic human study was published examining the dynamics of LH under continuous exposure to Kisspeptin-10.

The study included only three healthy adult men, Therefore, the results should be interpreted as a small physiological experiment, not as extensive clinical evidence.

The authors monitored the time course of LH and other hormonal parameters during a 24-hour experimental protocol.

Scientific interpretation: A more recent publication shows another characteristic of the human neuroendocrine response in a small experimental dataset. It does not represent a confirmed therapeutic or consumer function of Kisspeptin-10.

Level of scientific evidence

Data type What can be explored The main limitation

Genetic studies The importance of the endogenous KISS1/KISS1R system. They do not demonstrate the safety of an exogenous synthetic peptide.
Human physiological studies GnRH/LH dynamics in precisely defined protocols. The results cannot be automatically transferred outside of the specific protocol.
Research-grade COA Identity, purity and quantity of the specific batch. It does not demonstrate clinical safety or efficacy.

Regulatory and security context

The US FDA lists Kisspeptin-10 among the substances evaluated under the rules for bulk drug substances used in compounding.

In the regulatory assessment, questions related to, among other things, peptide materials are monitored:

  • immunogenicity,
  • peptide impurities,
  • characterization of the active pharmaceutical substance (API),
  • and the scope of available safety data.

Official source:

FDA – Certain Bulk Drug Substances for Use in Compounding that May Present Significant Safety Risks.

This regulatory process is not identical to the approval of Kisspeptin-10 as a medicinal product.

Research-grade Kisspeptin-10 and analytical documentation

The scientific publication describes the molecule used in a specific research protocol. It does not automatically provide information about the quality of a commercially available research-grade batch.

Therefore, the following are particularly relevant for the laboratory use of Kisspeptin-10:

  • the exact peptide sequence,
  • C-terminal amide addition,
  • molecular identity and mass,
  • chromatographic purity,
  • quantitative determination of quantity,
  • batch number or lot number,
  • the analytical method used,
  • the binding of COA to a specific analyzed sample,
  • Storage at –20 °C or 2–8 °C according to the label and the CoA of the specific batch.

The published analytical work using LC-MS/MS also showed that Kisspeptin-10 may be subject to degradation and the formation of shorter peptide products in biological matrices.

LC-MS/MS quantification and characterization of Kisspeptin-10 decomposition.

Analytical note: HPLC area purity, molecular identity, and the amount of peptide in a vial represent different analytical parameters. High chromatographic purity in itself does not confirm the correct amount, stability, or clinical safety of the material.

Storage of research material

Store Kisspeptin-10 of research grade at –20 °C or 2–8 °C according to the data indicated on the label and in the CoA of the specific batch. The material must remain in its original, properly labeled packaging and must be protected from conditions that may adversely affect its analytical integrity.

More about the evaluation of purity, COA and traceability of research-grade materials can be found in the article 7 criteria for selecting a peptide supplier.

The documentation available for the individual analyzed batches can be verified in the section COA and analytical documentation.

What does the available research not show?

It is extremely important not to translate the neuroendocrine mechanistic results of Kisspeptin-10 into consumer claims.

The available data by themselves do not show:

  • the suitability of Kisspeptin-10 for personal or human use,
  • the clinical safety of a particular research-grade material,
  • that the change in LH or GnRH signaling represents a universal health benefit,
  • that the results of healthy volunteers apply automatically to another population,
  • that the results of Kisspeptin-54 or KISS1R analogs automatically apply to Kisspeptin-10,
  • that the results of a particular clinical research material apply to a commercially available batch,
  • that the analytical purity of the product constitutes evidence of clinical safety or efficacy.

Therefore, it is not appropriate to use formulations such as „increases testosterone“, „supports fertility“, „regulates hormones“, „increases libido“ or other consumer-formulated hormonal benefits on the product or information page without precise experimental context.

How to critically read studies on Kisspeptin-10?

It is advisable to verify at each publication:

  • whether Kisspeptin-10, Kisspeptin-54 or a synthetic analogue of KISS1R was used,
  • whether it was a cellular, animal or human model,
  • which population was studied,
  • whether the result describes GnRH, LH, FSH or another parameter,
  • whether it is a one-time or continuous experimental protocol,
  • size of the studied dataset,
  • whether it is a primary study or an overview article.

Such a procedure helps to separate the biological mechanism from a claim that would go beyond the available evidence.
